I was thinking about how in Hurricane Fluttershy, all of the pegasi are required for the tornado. Rainbow Dash says that every pony should have ~10 wingpower, and they reach 800, though there are a few pegasi at home. So that makes just over 80 pegasi in Ponyville. And since it does not seem that any one kind of pony out-populates the others (with the exception of there being a million Colgates, of course), I estimated Ponyville's population somewhere around the 250 mark.
