The signal occurs in the round following Milner's first investment. If he chooses to try to add to (or defend) his ownership position in the NEXT financing round for one of the 42 companies he is sending a signal that only that one is interesting enough to warrant his attention. I'd assume he is trying to purchase an option to invest more money in the subsequent rounds by paying $150k to get an early look, establish a relationship and see how good the company/management team are. If he picks and chooses which companies he continues to put more money into he is tacitly rejecting the others, sending a negative signal to the other potential investors.
